PE-22-28 — Spadin-Derived Peptide for TREK-1 Channel Research
PE-22-28 is a synthetic heptapeptide from sortilin propeptide (spadin), selectively antagonizing TREK-1 potassium channels. Identified by Mazella et al. (PNAS, 2010). TREK-1 channels are highly expressed in hippocampus and cortex, regulating mood and cognition.
Mechanism of Action
PE-22-28 selectively blocks TREK-1 (KCNK2), increasing neuronal firing and synaptic transmission. TREK-1 knockout mice show enhanced hippocampal neurogenesis and BDNF signaling (Biological Psychiatry). PE-22-28 recapitulates these effects: increased neurogenesis, enhanced 5-HT neurotransmission, CREB/BDNF upregulation.
