Follistatin 344 (FST344) is a 344-amino acid glycoprotein that antagonizes activin, myostatin (GDF-8), and other TGF-β superfamily ligands. Research in FASEB Journal (2005) showed dramatic skeletal muscle mass increases in multiple animal models.
Mechanism of Action
Follistatin neutralizes TGF-β members through direct protein-protein interactions. Its three FS domains create a binding surface complementary to activin and myostatin receptor-binding epitopes, preventing ActRII/ALK4 activation and SMAD2/3 signaling. PNAS research showed sustained muscle mass increases in non-human primate gene delivery models.
