VIP — Vasoactive Intestinal Peptide for Neuroimmune and Mucosal Research
VIP is a 28-amino acid neuropeptide (Said & Mutt, 1970). Neurotransmitter and immune modulator acting through VPAC1/VPAC2 receptors across nervous, immune, endocrine, and GI systems. Over 10,000 published articles.
Mechanism of Action
VIP activates VPAC1/2 via Gs-cAMP signaling. In neurons: modulates synaptic transmission and circadian rhythm (key SCN synchronizer). In immune cells: generates tolerogenic DCs, promotes Tregs, shifts Th1→Th2 (Nat Rev Immunol). In gut: regulates ion transport, blood flow, smooth muscle.
Key Research Areas
- Neuroimmune Modulation: VIPergic nerves innervate lymphoid organs — studies how neuropeptide signaling regulates immune function.
- Circadian Clock Research: Essential for SCN neuronal synchronization. VPAC2 signaling maintains intercellular clock coupling.
- Regulatory T-Cell Biology: Promotes FoxP3+ Treg differentiation — peripheral tolerance and immune homeostasis mechanisms.
- Gut-Brain Axis: Abundant enteric neuropeptide regulating secretion, motility, barrier function, and mucosal immune responses.
